Chemokines
Chemokines constitute a super-family of small, inducible, secreted, pro-inflammatory cytokines involved in a variety of immune responses, acting primarily as chemoattractants and activators of specific types of leukocytes. Four classes of chemokines have been defined by the arrangement of the conserved cysteine (C) residues of the mature proteins:
Chemokines mediate their activities by binding to target cell surface chemokine receptors that belong to the large family of G protein-coupled, seven transmembrane domain receptors. CSS uses novel technologies developed for the synthesis of long peptides to chemically synthesise members of the chemokine family. It uses special folding conditions to produce biologically active chemokines and has now synthesised and purified more than 20 wild type and modified chemokines. Chemical synthesis allows the production of chemokines that are very difficult, if not impossible, to produce recombinantly. It also opens up the opportunity to site specifically modify the wild type, eg to improve the design of potential therapeutic candidates or to fluorescently label or biotinylate chemokines for a variety of applications including assay development, diagnostics and basic research.
